CircINSR Regulates Fetal Bovine Muscle and Fat Development

چکیده
The level of muscle development in livestock directly affects the production efficiency livestock, and contents intramuscular fat (IMF) is an important factor that meat quality. However, molecular mechanisms through which circular RNA (circRNA) IMF remains largely unknown. In this study, we isolated myoblasts preadipocytes from fetal bovine skeletal muscle. Oil Red O BODIPY staining were used to identify lipid droplets preadipocytes, anti-myosin heavy chain (MyHC) immunofluorescence was myotubes differentiated myoblasts. Bioinformatics, a dual-fluorescence reporter system, pull-down, RNA-binding protein immunoprecipitation determine interactions between circINSR micro (miR)-15/16 family. Molecular biochemical assays confirm roles played by preadipocytes. We found able differentiate normally. CircINSR serve as sponge for miR-15/16 family, targets CCND1 Bcl-2 . overexpression significantly promoted myoblast preadipocyte proliferation inhibited cell apoptosis. addition, adipogenesis alleviating inhibition against target genes FOXO1 EPT1 Taken together, our study demonstrated serves regulator embryonic development.

Phosphodiesterase 9 (PDE9) regulates bovine tracheal smooth muscle relaxation
The present study was designed to clarify phosphodiesterase 9 (PDE9) expression in bovine tracheal smooth muscle tissue, and to elucidate that PDE9 may contribute to the regulation of airway relaxation. PDE9 mRNA expression was detected in bovine tracheal smooth muscle. Sodium nitroprusside (an NO donor) and BAY 73-6691 (a selective PDE9 inhibitor) reduced high K+- and carbachol-induced contrac...
متن کاملNfix Regulates Fetal-Specific Transcription in Developing Skeletal Muscle
Skeletal myogenesis, like hematopoiesis, occurs in successive developmental stages that involve different cell populations and expression of different genes. We show here that the transcription factor nuclear factor one X (Nfix), whose expression is activated by Pax7 in fetal muscle, in turn activates the transcription of fetal specific genes such as MCK and beta-enolase while repressing embryo...
متن کاملIUGR alters muscle fiber development and proteome in fetal pigs.
Intrauterine growth restriction (IUGR) may have permanent stunting effects on muscle growth and development of the progeny. However, underlying mechanisms are largely unknown. Recent studies comparing muscle fiber development and proteomes in IUGR and normal-body-weight (NBW) fetal pigs indicated that muscle fiber diameter were smaller in IUGR fetal pigs than in NBW fetal pigs on all three stag...
متن کاملDevelopmental Programming of Fetal Skeletal Muscle and Adipose Tissue Development
All important developmental milestones are accomplished during the fetal stage, and nutrient fluctuation during this stage produces lasting effects on offspring health, so called fetal programming or developmental programming. The fetal stage is critical for skeletal muscle development, as well as adipose and connective tissue development. Maternal under-nutrition at this stage affects the prol...
متن کاملSertoli Cell Wt1 Regulates Peritubular Myoid Cell and Fetal Leydig Cell Differentiation during Fetal Testis Development
Sertoli cells play a significant role in regulating fetal testis compartmentalization to generate testis cords and interstitium during development. The Sertoli cell Wilms' tumor 1 (Wt1) gene, which encodes ~24 zinc finger-containing transcription factors, is known to play a crucial role in fetal testis cord assembly and maintenance. However, whether Wt1 regulates fetal testis compartmentalizati...
